I'm setting up an Asset Builder page for pdf files, and associated Asset Listing page to autolist pdf documents. I would like to include metadata fields (eg caption) that would allow each file listing to include some information about the file. The Title (friendly name) isn't sufficient.
Can one specify metadata fields (eg %asset_metadata_comment%) within the Asset Builder for the user to fill in? If so, in which version ?

No, you can't. The asset builder only presents the default create screen.
You'd need to create the asset first, using the asset builder, and then redirect the user to the simple edit interface for the newly created asset. You can print the metadata inteface there for them to fill in.

This is exactly what I want to do!
And I also tried the redirect to the simple edit interface (http://adventist.org.au/__data/assets/pdf_file/26221/Part_2.pdf/_edit) but it comes up with “Page not found”.

The Live URL for assets (i.e. any URL with __data) cannot have a simple edit interface (as this is served directly by Apache).
You need to get the Matrix URL for the file and add /_edit to that.

You do this by creating a layout for the asset type. Layouts define the simple editing interface for assets. On the layout, use the keyword to print the metadata screen. The system admin manual will have more info for you.

What is the best way of getting this url avi… tried the system URL, with a=xxx/_edit and still got a 404…
You can use the URL of http://www.url.com/_edit?limbo_assetid=XXX to get to the Simple Edit interface for live, File-based assets.

on each of the site assets however, there is a http://outernode.pir.sa.gov.au/
